If used up rather quickly within weeks, mostly refrigerated temperatures are fine, but -20°C is preferred. When stored for months and years, store them at -20°C or ideally at -80°C. After longer storage, ideally test purity through analytical HPLC-MS.
Peptide Libraries
We start to call it libraries from 48 peptides onwards, but this term is a vague definition.
We can essentially any scale in the mg range in library format, but the regular amount would be 1-2 mg per peptide. We use automated synthesizers, that can generate many peptides in parallel in that scale.
Yes, we surely can synthesize peptide libraries with chemical modifications.
Very often, peptides ordered in library formats are used in early discovery stage and thus, very high purities may not be required. They may often be used as crude material without purification or at a guaranteed purity of above 70%. However, also higher purities are possible.
When screening for MHC class I T cell epitopes, often 9-11mer peptides are used, which should be synthesized with a C-terminal acid. For MHC class II T cell epitope and B cell epitope screenings, C-terminal amides work as well and are more robust than acids.
Peptides are delivered as freeze-dried powder in separate vials, except for different arrangements discussed prior to the synthesis.
